Active substances in cereal products for baby food

Present bachelor thesis is focused on the study of antioxidant substances in cereal products for baby food. The theoretical part is aimed at describing the anatomical structure and chemical composition of cereal grain, characterization of antioxidants with the focus on natural antioxidants, the technology and nutritional parameters of cereal products. The experimental part deals with the content of phenolic compounds, flavonoids, and the total and reducing sugars in the total of 12 kinds of cereal products with different flavour – 5 types of oak pap, 3 types of fiber pap and 4 corn paps. The work includes the analysis of ascorbic acid by titration method and the analysis of tocopherol, carotenoids and catechins by high performance liquid chromatography as well. The highest content of total phenolics and flavonoids was found in corn pap with raspberries and also in cereal pap with chocolate. High levels of vitamins E and C were found predominantly in flavoured fiber pap.
Characterization of cereal products with fruit component for baby nutrition

Present bachelor thesis deals with the characterization of milk cereal products intended for baby food. The theoretical part focuses on the morphology of cereal grains, fortification, an overwiev of supplementary components used to fortification and their properties. For the practical part eight kinds of cereal dairy products of three different producers were chosen. In these products total polyphenols, total flavonoids, reducing and total saccharides and proteins were analyzed. The highest level of polyphenols was measured in product Sunar multicereal. The highest amount of flavonoids (more than 50 % of total phenolics) was found in Nestle fruit product. The highest level of total and reducing saccharides contained Nestlé honey flavour. High amount of proteins was found in products Sunar with multicereal flavour and pears with biscuits. Using HPLC/UV-VIS analysis vitamins C and E were determined. The highest content of vitamin C contained Nestlé with cookie lavor, while high vitamin E level was found in Sunar pear flavor and biscuits. Using HPLC/RI individual saccharides were examined. In all products high amount of lactose was found, mainly in the products of Sunar. Nestlé products contained mostly glukose, while in products Hami sucrose was detected as the most abundant saccharide.
The application of magnetic particles for DNA isolation from cereal products

The thesis has been focused on micro method for isolation of PCR- ready DNA iusing magnetic particles from cereal products. Cereal biscuits and cereal products for babies were selected for the analysis. These were homogenized using plastic copist in lysis buffer with cetyltrimethylammonium bromide (CTAB). The homogenates were purified using chloroform- octanol mixture. The effect of isopropanol in the preparation of homogenates was tested, too. Homogenates were used for DNA isolation by magnetic particles. Two ways to isolate magnetic particles with bounded DNA (magnetic separator and magnetic needle have been tested. Isolated DNA was analyzed spectrophotometrically its concentration and purity were assessed. . After that, amplification of the DNA was tested in PCR. Two sets of primers specific for plant ribosomal DNA were used for their amplification. PCR products of expected length 700 bp and 220 bp were detected by agarose gel electrophoresis. It was shown that DNA isolated from seeds and cereal products using magnetic particles was in PCR-ready quality.

Representation of cereal products and their quality in the diet of basic school pupils and university students

In the theoretical part of the diploma thesis there was described food of cereal character including school nutrition programmes and their representation in the diet of basic school pupils and university students. The interview focused on the representation of cereal products and their importance in the diet is described in the research part of this diploma thesis. The interview was made in the groups of pupils in higher basic schools and university students. When evaluating the questionnaire survey, there were assessed the quantity, individual types and quality of commonly consumed cereal products and the level of eating habits of respondents in accordance with the principles of good nutrition. After evaluation of the interviews it was found that basic school pupils have worse all-day eating habits than university students. It was confirmed mostly by the answers of the questions relating to breakfast, mid-afternoon snacks and total number of meals during a day. University students prefer wholegrain products more than basic school pupils. Pupils of higher basic schools have lower knowledge level of cereals than university students.
Analysis of the cereal food products using mathematical modelling

The bachelor thesis deals with nutritional values of cereal food products from three companies (Nestlé, Bona Vita and Emco). In the first part of the thesis linear programming models are described. The next chapter deals with the characteristics of the real situation in which Guideline Daily Amounts and nutritional values of specific cereal food products are set, and economic and mathematical model is formulated. The last part is devoted to numerical experiments. At first, the models are solved in MS Excel 2007 and then some of them are analysed in LINGO modelling system. The main aim of the thesis is to determine whether cereal food products are healthy and nutritious in terms of their nutritional values. The proposed models determine the individual portions of cereal food products consumed per day using the minimum costs that are spent for the purchase of the products.
